Researchers at the Fred Hutchinson Cancer Research Center, Seattle, were trying to find an answer to the puzzling question why cancer cells were easy to kill under laboratory conditions but hard to eradicate once growing inside the human body (a typical chemotherapy treatment will initially show good tumor response, but will be followed by rapid regrowth and eventual resistance to further [chemotherapy] treatment).
In the course of their testing the effects of a certain type of chemotherapy on prostate (and later breast and ovarian) cancer tissue, the scientists made a surprise discovery: healthy cells apparently sustained DNA damage from the chemotherapy treatment, which led to increased secretion of a protein named WNT16B. This protein was taken up by the neighbouring cancer cells, enhancing their growth and invasive behaviour and most importantly making them resistant to subsequent (chemo) treatment.
The conclusion: in addition to its general extreme toxicity (compare e.g. High risks involved in accidental spillage of chemotherapy drugs), one major reason why chemotherapy is likely to make cancer more aggressive is the fact that it inflicts a specific type of damage on healthy cells expressed as increased secretion of a protein (WNT16B) which boosts cancer cell survival.
The above study (Treatment-induced damage to the tumor microenvironment promotes prostate cancer therapy resistance through WNT16B)—funded inter alia by the NIH (National Institutes of Health) and the NCI (National Cancer Institute) of the USA—was published in Nature Medicine in 2012.
